What the human trials actually show
This is where the honesty matters. The flagship human study gave 3 grams of glycine before bed to people with ongoing sleep complaints and measured them with polysomnography. Glycine shortened the time to fall asleep and the time to reach slow-wave sleep, and improved subjective sleep quality, without distorting overall sleep architecture.[5]
A separate trial restricted healthy volunteers to 25 percent less sleep for three nights and gave 3 grams of glycine before bed. Glycine reduced daytime fatigue and sleepiness, with the clearest benefit on the first sleep-restricted day.[3]
Then comes the cold water. A 2024 systematic review that pooled the human glycine literature concluded that while longer-term glycine did improve sleep in healthy people, the studies were small and carried a high risk of bias, with only a minority of trials rated as low-risk.[4] So the direction of effect is consistent across the trials, but the quality of evidence is not strong enough to call it settled.
